You wouldn't need to, since you already have those notes using a combination of black and white keys. The user scale feature is if you are doing eastern music or experimental music that uses notes not on a piano. If there was less than 1/2 step between notes. Or, for doing classical music with popular tunings of the time such as just intonation, or pythagorean intonation (these would only sound good in one key though).

Auto-lock, you will need to detune your B to -50 cents. Go to the Global area of the Radias and navigate to user scale then select B and go to -50cents.

In the program, select user scale.
