Keyboard Part help on ESX-1

Hi there,
I am having a little trouble making synth patches / Keyboard parts on my ESX1
I know how to load a sample to the keyboard part but how do I get it in tune so when you play the note on the keys your base note is in C therefore when playing the next note it will be a D, E etc etc working like a proper synth??
Is the trick to tune the sample to C before you load it into the ESX1. I am trying to create those long sustained leads sounds that work so well with the appg mode reversed but i am getting a little bored of the factory synth parts.
And what kind of of samples would be best to use? The factory samples work so well as synth parts. I am just not sure how to go about making these kind of synth patches.
If this makes sense to you some advice would be fully appriciated.
