You have to save the sample(s) in Media Mode.
When you go to media and click the save tab, in the upper right hand corner is a place to save pcg, seg, etc... Click save .ksc as well as seq or whatever you plan on saving (.mid, smf, etc.).
This will save all the changes you made to any samples.

I've done that, I've saved the sample to media, but when I turned it off and turned it on, the program I made (sample converted into program) was not saved. And I have to load the sample again from the media and convert it again to program.
My question is how do I save the program I made that came from sample, like when I turned off and on, the program (converted from sample) is still there without loading the sample file from the media, is this possible?

You are probably missing some of the steps. When you make your multisample and convert it to program, you have to internally write that program (option in upper right dropping menu in program mode), then go to media mode and save .pcg and sampling (.ksc) data or just choose to save all.

I see, I'll double check my procedures, I think I missed the saving of PCG/ all files to media, thanks shrike!

I did forget to mention that when you turn on the Triton, you need to load those .ksc files along with your sequence.
