yep, I get it too, on every post. Then it must stop something else because my posts show up as unread and I have to go and read what I've just written (kind of annoying)

I only get it on some threads, not all. I didn't get it on this one for example.
My guess is someone has ticked "Notify me when a reply is posted" but not entered a working email address. Just a guess though

well, i have that on automatic. I didn't get it when I posted here. So it must be my post that's activated the mailer function.
However, I can assure you that my mail is working fine.
EDIT:
and I didn't get it posting this either, which makes sense because you don't get notification e-mails about your own replies.
However, I've experienced this on other threads, so its certainly not my e-mail address.

Damn, I thought it was just me. Just started happening yesterday. I think I know the problem and will fix very soon !

My first attempt to fix didn't work.
X-Trade's note gave me a useful clue. The error means the our email server (SMTP) configuration is not correct. Or rather it was correct but yesterday our hosts must have changed something and broke our configuration.
If you are seeing this error, mailing from the server has not been properly configured. The error will be displayed anytime that an email would be sent (right after registration, after posting, after sending a PM, etc.).
I am off down in the engine room to smack things around

It should be fixed now. Thanks guys for the spot, let me know how it is working for you. Post here, because I have the reply notification enabled and that was the piece failing.
